Electron capture gas-liquid chromatographic-mass spectral identification of acids produced by Neisseria meningitidis in a defined medium.

چکیده
The acid metabolites and the cellular fatty acids of three strains of Neisseria meningitidis grown in a chemically defined liquid medium were determined with computerized frequency-pulsed electron capture gas-liquid chromatography. Five acids not previously reported were subsequently identified: isobutyric, octanoic, decenoic (C10:1), dodecenoic (C12:1), and tetradecenoic (C14:1). These acids were produced during active metabolism and were not detected as cellular constituents. The frequency-pulsed electron capture gas-liquid chromatography methods which we used provide a rapid, reliable, sensitive means of detecting both these and other metabolic and cellular acids in spent culture medium.

Identification of Neisseria by electron capture gas-liquid chromatography of metabolites in a chemically defined growth medium.
A dual-purpose study was carried out in an attempt to develop a rapid, sensitive method to identify Neisseria species by gas chromatography and to learn more about the metabolism of these organisms. Sixty-nine isolates of Neisseria were grown in a chemically defined fluid medium; the spent medium was extracted sequentially at pH 2 with diethyl ether and at pH 10 with chloroform. The pH 10 extra...
متن کاملCellular fatty acids of pathogenic Neisseria.
The cellular fatty acid composition of 20 isolates of Neisseria gonorrhoeae and 21 isolates of N. meningitidis was examined by gas-liquid chromatography. Each isolate of the two species possessed similar fatty acid profiles which were characterized by five major acids, accounting for 80 to 85% of the total. The three most abundant acids in each species were palmitic, palmitoleic, and beta-hydro...
متن کاملAn electron-capture gas-liquid-chromatographic method for the determination of prostaglandin F2alpha in biological fluids.
A sensitive electron-capture gas-liquid-chromatographic method for the determination of sub-nanogram quantities of prostaglandin F(2alpha) was developed. The method is based on the sub-microgram scale conversion of the prostaglandin into the electron-capturing pentafluorobenzyl ester, and analysis of the latter as the tris-trimethylsilyl ether. The lower limit of detection was 12.5pg of the est...
متن کاملAqueous in situ derivatization of carboxylic acids by an ionic carbodiimide and 2,2,2-trifluoroethylamine for electron-capture detection.
We report a technique for the rapid, room temperature derivatization of aqueous carboxylic acids to the corresponding 2,2,2-trifluoroethylamide derivative. 3-Ethyl-1-[3-(dimethylamino)propyl]carbodiimide hydrochloride (EDC) and 2,2,2-trifluoroethylamine hydrochloride (TFEA) were added to aqueous samples of several acids of interest in environmental analytical chemistry, including benzoic acid, ...
متن کاملDevelopment of a DNA Aptamer for Screening Neisseria meningitidis Serogroup B by Cell SELEX
Background: Artificial oligonucleotides like DNA or RNA aptamers can be used as biodiagnostic alternatives for antibodies to detect pathogens. Comparing to antibodies, artificial oligonucleotides are produced easily at lower costs and are more stable. Neisseria meningitidis, the causative agent of meningitis, is responsible for about 1% of infections in an epidemic period. Specific DNA aptamers...
